OHSU # 2532 — Virtual patient population for mathematical representation of glucose metabolism in type 1 diabetes

Summary

A method for in silico validation and development of devices and algorithms for glucose management, such as insulin pumps.

Technology Overview

This is a mathematical model of the glucoregulatory system.  It is comprised of a set of differential equations that represent compartments in the human body through which glucose is metabolized.  The inputs to the model are carbohydrates, exercise, insulin, and glucagon.  The output of the model is plasma glucose.  From this, a person’s plasma glucose level can be estimated based on food intake as well as drug usage (insulin and glucagon) and physiologic interventions such as exercise. The OHSU VPP has been validated against actual clinical data in patients with type 1 diabetes undergoing closed loop insulin and glucagon therapy.  The virtual population has been compared with a population of real patients who are close matches physiologically with true patients with type 1 diabetes.

Using this model, any number of in-silico (virtual) patients can be created by varying the model parameters such as insulin sensitivity, carbohydrate sensitivity, the insulin kinetics time constant, and the carbohydrate kinetics time constant to run physiology tests prior to running actual human studies.
